3CL Protease Inhibitor Herb for Fighting Infections

Amazon may not be the best place, but you are looking for the gromwell root extract or the root powder (not leaves), or Gromwell Dried Bulk Herb, or Root: Lithospermum Root (Gromwell Root) / Ja Cho Geon 3oz

This is all new information to me, so I haven’t investigated the use of the $20 herbal extract vs the $99+ Tollovid. The main idea is that this Chinese herb is a protease inhibitor and very effective in dealing with the (xxx) sickness.

Lithospermum erythrorhizon (also known as purple gromwell or zicao / zi cao ) is the plant. The dried root contains Shikonin, a Chinese herbal medicine with various biological activities, including inhibition of human viruses [source].

How to buy and use Gromwell Root

Often, Gromwell root is sold as a powder or dried root. The powdered root may be a simpler to use for beginners. Recommendation from The Alchemist’s Kitchen herbalists: “To use internally, mix 1 tsp of Gromwell root powder with hot water, tea, broth, liquid, or food of choice. The powder can also be encapsulated in vegetable cellulose capsules and taken as a supplement. Alternatively, the powder can be mixed with vinegar, water, tea, aloe, honey, or liquid of choice. It can also be applied as a poultice to wounded or inflamed skin. The powder can be added to face oils or creams or used as a face mask.”

From the seller: If you are using powdered Gromwell Root Powder or any other powdered root, always start slowly and ONLY make a dilute infusion because you do not know your reaction to the herb. Whether it is a single herb or a mixture, try only one teaspoon of herb per 16 oz of water at first. Monitor your body’s reaction, feeling better-the same-worse, and if the desired response occurred, increase the amount to tablespoon and monitor your body’s response. 

Tea brewing method
1. Rinse the herbs with cold running water then soak it for 30 min.
2. Combine 1 quart Water : 1 Tbs Herbs
3. Bring to a boil, lower the heat and simmer for 30 min to 2 hr

Direction for infusion or tisane.

Protease inhibitors influence a range of innate immunity and inflammatory pathways.

Severe (xxx-VID) is characterized by inflammatory and immuno-thrombotic pathway activation, many of which are regulated by protease inhibitors.

Alpha-1-antitrypsin (AAT) is a serine protease inhibitor (SERPIN) and the third most abundant protein in circulation. 

Because alpha-1-antitrypsin (AAT) embraces a panoply of biologic activities that may antagonize several pathophysiologic mechanisms induced by SARS-CoV-2, we hypothesize that this naturally occurring molecule is a promising agent to ameliorate XXX-VID-19.

We posit at least seven different mechanisms by which AAT may alleviate XXX-vid-19.

  • First, AAT is a serine protease inhibitor (SERPIN) shown to inhibit TMPRSS-2, the host serine protease that cleaves the spike protein of SARS-CoV-2, a necessary preparatory step for the virus to bind its cell surface receptor ACE2 to gain intracellular entry.
  • Second, AAT has anti-viral activity against other RNA viruses HIV and influenza as well as induces autophagy, a known host effector mechanism against MERS-CoV, a related coronavirus that causes the Middle East Respiratory Syndrome.
  • Third, AAT has potent anti-inflammatory properties, in part through inhibiting both nuclear factor-kappa B (NFκB) activation and ADAM17 (also known as tumor necrosis factor-alpha converting enzyme), and thus may dampen the hyper-inflammatory response of xxx-VID.
  • Fourth, AAT inhibits neutrophil elastase, a serine protease that helps recruit potentially injurious neutrophils and implicated in acute lung injury. AAT inhibition of ADAM17 also prevents shedding of ACE2 and hence may preserve ACE2 inhibition of bradykinin, reducing the ability of bradykinin to cause a capillary leak in xxx – vid.
  • Fifth, AAT inhibits thrombin, and venous thromboembolism and in situ microthrombi and macrothrombi are increasingly implicated in xxx- VID.
  • Sixth, AAT inhibition of elastase can antagonize the formation of neutrophil extracellular traps (NETs), a complex extracellular structure comprised of neutrophil-derived DNA, histones, and proteases, and implicated in the immunothrombosis of xxx – VID; indeed, AAT has been shown to change the shape and adherence of non-xxx-VID-related NETs.
  • Seventh, AAT inhibition of endothelial cell apoptosis may limit the endothelial injury linked to severe xxx – VID-19-associated acute lung injury, multi-organ dysfunction, and pre-eclampsia-like syndrome seen in gravid women.
  • Furthermore, because both NETs formation and the presence of anti-phospholipid antibodies are increased in both xxx – VID and non-xxx – VID pre-eclampsia, it suggests a similar vascular pathogenesis in both disorders. As a final point, AAT has an excellent safety profile when administered to patients with AAT deficiency and is dosed intravenously once weekly but also comes in an inhaled preparation. Thus, AAT is an appealing drug candidate to treat xxx – VID and should be studied.


A total of 57 phytochemicals were scrutinized virtually against four structural protein targets of SARS-CoV-2 viz. 3CLpro, ACE-2, spike glycoprotein and RdRp. Providentially, two bioactives from each of the three plants i.e. apigenin-o-7-glucuronide and ellagic acid from Eucalyptus globulus; eudesmol and viridiflorene from Vitex negundo and; vasicolinone and anisotine from Justicia adhatoda were identified to be the best hit lead molecules based on interaction energies, conventional hydrogen bonding numbers and other non-covalent interactions. On comparison with the known SARS-CoV-2 protease inhibitor –lopinavir and RdRp inhibitor –remdesivir, apigenin-o-7-glucuronide was found to be a phenomenal inhibitor of both protease and polymerase, as it strongly interacts with their active sites and exhibited remarkably high binding affinity. Furthermore, in silico drug-likeness and ADMET prediction analyses clearly evidenced the usability of the identified bioactives to develop as drug against xxx – VID.


Overall, the data of the present study exemplifies that the phytochemicals from selected traditional herbs having significance in steam inhalation therapy would be promising in combating xxx – VID.

Lithospermum erythrorhizon

Lithospermum erythrorhizon (LE) has also been used in traditional medicine for its various pharmacological effects.

LE contains naphthoquinones, including shikonin and its derivatives, identified as the major phytochemicals. Recent studies have reported the various biological activities of LE, including

  • anti-inflammatory, antiobesity, and antilipogenic activities [2830].
  • LE is a complementary herb that can act simultaneously on various targets in diseased states.
